BI102 General Biology Spring 2008
|
Dr. Brian K. Penney |
Plant evolution and life cycles will be introduced 31 Jan, so all students should be familiar with this material. Plant structure (the bulk of this lab) will be covered 5 Feb, so Monday and Tuesday labs will need a bit more explanation.
LAB SCHEDULE
INTRO (10 min): 1 Plant cells and tissue systems
2 Plant organs (30 min)
A Leaves
B Roots
C Stems: Instructors make slides as demo; changes: using wandering jew (Zebrina) instead of Coleus. STUDENT DRAWINGS OF STEMS MUST INCLUDE EACH TISSUE TYPE (color coded, or whatever) FOR TWO POINTS ON QUIZ. You will have to INITIAL their quiz. Drawings should indicate the overall cross section, plus what cell types look like in each tissue system covered.
3 Dermal tissue (15 min)
A in stems
B in roots
C in leaves NO TISSUE PEEL but look at leaves under scope to see guard cells
4 Ground tissue (15 min)
A Parenchyma: stem XS ONLY
B collenchyma: stem XS ONLY
C sclerenchyma (Pear slices)
5 Vascular tissue (10 min)
A in leaves SKIP
B in roots: students should see vascular cylinder
C in stems
DEMO/DISCUSSION (20 min): grocery store botany: identify whether "vegetable" is fruit, root, stem, leaf
DEMO/DISCUSSION (10 min): Students observe fungal hyphae and structure.
QUIZ (10 min)
Done as a demonstration so students understand how histological sections are made. Stems will NOT be embedded in paraffin in nut and bolt microtome as indicated in lab manual; instructors will simply do thin slices of fresh stem for students to see.
Required Equipment: A Staining histological sections| Type | # | Per | Item | Section | Notes |
|---|---|---|---|---|---|
| E | 1 | Class | razor blade | A Staining histological sections | |
| E | 1 | Class | blank slide and cover slip | A Staining histological sections | |

Students look at slides, draw overall cross sections and major features.
Required Equipment: B Plant organs| Type | # | Per | Item | Section | Notes |
|---|---|---|---|---|---|
| E | 1 | Pair | Leaf cross section slide | B Plant organs |
|
| E | 1 | Pair | Root cross section slide | B Plant organs |

|
| E | 1 | Pair | Compound microscopes | B Plant organs | |
| L | 1 | Class | germinating seeds | B Plant organs | pre-sprout on moistened paper towel |
Students go back over the same slides they have already observed, this time to see where various tissue systems appear in each cross section.
Here is a diagrammatic view of tissue systems among various plant organs, from text figure 31.6
| Type | # | Per | Item | Section | Notes |
|---|---|---|---|---|---|
| 1 | slides as for Plant organs exercise | C Plant tissue and cell types | no changes, students just view them again with different focus |

| E | 1 | Pair | pair foceps | C Plant tissue and cell types |
GOAL: students understand the plant parts that comprise typical food items
Required Equipment: D Grocery store botany| Type | # | Per | Item | Section | Notes |
|---|---|---|---|---|---|
| L | 1 | Class | various leaf vegetables | D Grocery store botany | cabbage, lettuce, celery (petiole/leaf) |

| L | 1 | Class | various stem vegetables | D Grocery store botany | broccoli (technically stem and flowers), asparagus |
Students should view slides of fungal hyphae to compare to plant morphology, and understand how they differ.
